    BioPhi outputs and interpretation

Humanness scores, classification, humanized sequences, and mutations from BioPhi.

    biophioutputs

BioPhi returns an OASis humanness score and percentile, a human/humanized/chimeric/murine classification, and a per-CDR and per-framework breakdown. In a humanization run it also returns the rewritten VH/VL sequences and the list of mutations it made. Read the classification and the per-region breakdown together: a merely "humanized" class with a low-humanness framework region points you straight at where more work is needed.

    Humanness fields

    FieldTypeWhat it tells you
    humanness_scorefloatOverall OASis humanness (0–1).
    oasis_percentilefloatOASis percentile rank (0–100).
humanness_classhuman | humanized | chimeric | murineClassification from the OASis percentile.
    vh_humannessfloatHeavy-chain humanness.
    vl_humannessfloat | nullLight-chain humanness.
    cdr_humannessmap region→floatPer-CDR humanness (e.g. CDR-H1).
    framework_humannessfloatCombined framework-region humanness.
    immunogenicity_risklow | moderate | highPredicted immunogenicity risk.

    Humanization fields

Present when the job ran in a humanization mode:

    FieldTypeWhat it tells you
    humanized_vhstring | nullHumanized heavy-chain sequence.
    humanized_vlstring | nullHumanized light-chain sequence.
    mutations_madelistMutations applied, each as { position, from_aa, to_aa }.
immunogenicity_reduction_percentfloat | nullEstimated immunogenicity reduction from humanization.

    Reading the result card

The BioPhi result renders on the canvas as a humanization card, surfacing the humanness score, class, and (in humanization mode) the mutations so you can see the change at a glance.

    What to do next

    • Feed the humanized_vh/humanized_vl into antibody developability — humanization can move charge and aggregation.
    • Re-number the humanized sequence with ANARCI.
    • Confirm the mutations avoid the paratope so binding is preserved.
