Humatch quickstart
Paste a sequence, set the chain to H or L, and read back a humanness score, the closest human germline V gene, and the low-humanness positions to target.
You have an antibody sequence and you want a humanness read — an overall score, the least-human positions, and the closest human germline. Humatch does it from sequence alone: paste the sequence, tell it whether the chain is heavy or light, submit, and read the score and flagged positions off the result card.
Score a sequence
Provide sequences
Paste one or more antibody variable-region sequences (up to 50 per job). Name them if you want the labels carried through.
Set the chain type
Choose H for heavy chains or L for light (kappa or lambda). Keep the default imgt numbering scheme unless a downstream convention needs Chothia or Kabat.
Submit and wait
Humatch runs as an asynchronous CPU job on Modal. Submit it, and a result card appears on the canvas when the job finishes — typically within two to three minutes.
Read the score
The card shows each sequence's humanness score, whether it passes your threshold, the closest human germline V gene, and the flagged low-humanness positions to target for humanization.
In chat
Describe the antibody and ask for a humanness read — the model routes to Humatch and pairs it with numbering or annotation when your prompt asks.
What to do next
- Humanize the flagged positions — feed the sequence into BioPhi for Sapiens humanization or CDR grafting.
- Check manufacturability — run antibody developability.
- Map the paratope — run Paragraph so you humanize away from binding residues.
